The aim of the project is development of novel methods for detecton of DNA damage resulting from exposure to polyaromatic hydrocarbons (PAH). Initially, PAH-DNA aducts will be prepared under enzymatic catalysis of cytochrome P450 and using chemical synthesis. The aducts will be used to rise specific antibodies (monoclonals as well as polyclonals) which will further serve for development of immunochemical methods including ELISA (standard, nanoparticle- and enzyme recyclation-amplified variants, opticaland electrochemical detection) and immunosensors (surface plasmon resonance system Biacore, piezoelectric resonators). Biosensor-based technologies will also serve for detailed characterization of the newly prepared bioreagents. The localization of DNA damage will be studied using atomic force microscopy, nanoparticle-labeled antibodies will serve for visualization of the point of damage. Finally, the newly developed detection methods will be tested on real samples and validated with alternative approaches.
