Head and neck squamous cell carcinoma (HNSCC) is common and severe solid cancer. Cancer-associated fibroblasts (CAFs) can promote HNSCC progression and therapy resistance by secreting factors contained in exosomes. Exosomes are released from CAFs and other cells in the tumour microenvironment and may increase the invasive behaviour of cancer cells. Exosomal proteins are promising non-invasive biomarkers for many types of cancer as exosomes with tumour-specific proteins can be also detected in the venous blood of cancer patients. In this project, we want to investigate the proteome of CAFs-derived exosomes. We will test the relevance of exosomal proteins as biomarkers for diagnosis, relapse-free survival, and treatment resistance of HNSCC patients indicated for curative surgery and adjuvant chemotherapy. Furthermore, we would like to assemble a set of CAFs-derived exosomal proteins that can be detected also in the venous blood of HNSCC patients and reflect the presence of a malignant tumour or treatment-resistant phenotype.
